چکیده

Various oncogenes correlate with tumor phenotypes, which represent transformed morphology, loss of contact inhibition, colony formation in soft agar and tumorigenicity in nude mice. Thus, inducers of phenotypic reversion in oncogene-transformed cells mayact as selective antitumor agents without cytotoxicity. Our screening for such substances resulted in the isolation of trichostatin D (Fig. 1), a new member of the trichostatin family12), from an actinomycete identified as Streptomyces violaceusniger. This compound induced flat reversion in NIH3T3murine fibroblasts transformed with human papillomavirus oncogenes (NIH3T3/T-601)3). We report here the

Morphological reversion of sis-transformed NIH3T3 cells by trichostatin A.

Trichostatin A (TSA) induced the normal and flat phenotype of sis-transformed NIH3T3 cells at quite a low concentration of 1 ng/ml. Although morphological changes were found in other oncogene-transformed cells, they were not the same as those seen for the sis-transformed cells. Almost complete reversion into the flat phenotype was seen at 6 h after administration of the compound, suggesting tha...

Trichostatin A Promotes the Conversion of Astrocytes to Oligodendrocyte Progenitors in a Defined Culture Medium

The generation of oligodendrocyte progenitor cells (OPCs) offers tremendous opportunities for cell replacement therapy in demyelinating diseases such as multiple sclerosis (MS) and spinal cord injury. Recently, the prospect of reprogramming terminally differentiated adult cells towards another mature somatic cell or progenitor cells without an intermediate pluripotent state has been of interest...

Depudecin induces morphological reversion of transformed fibroblasts via the inhibition of histone deacetylase.

Depudecin is a fungal metabolite that reverts the rounded phenotype of NIH 3T3 fibroblasts transformed with v-ras and v-src oncogenes to the flattened phenotype of the nontransformed parental cells. The mechanism of detransformation induced by this agent had not been determined. Here, we demonstrate that depudecin inhibits histone deacetylase (HDAC) activity effectively both in vivo and in vitr...

DNA-mediated alteration of the reversion frequency of transformed NIH/3T3 cells.

Cell selection immediately after DNA-mediated transfection of whole-cell DNA into mammalian cells has been used to select for specific DNA sequences that cause a phenotypic effect. Whole-cell mouse or human DNA was cleaved into a distribution of lengths (0.4-25 kilobase pairs) and transfected into anchorage-independent spontaneously transformed NIH/3T3 cells. Immediately after transaction, anch...
